Fruhvirtova was given a wildcard into the Miami Open qualification for 2021, but she was defeated in the first round by Nina Stojanovic. She received a wildcard into the main event of the WTA 250 MUSC Health Women’s Open in April when she defeated Alize Cornet in the third set to claim her maiden WTA singles victory. The Czech then proceeded to the quarterfinals by defeating Emma Navarro in straight sets in the next round, however she was defeated in the final by eventual champion Astra Sharma.
