Defending Champion Madison Keys is seeded eighth for this event. She missed the whole Australian Summer because she had tested positive for Covid. Her first tournament back was the Qatar Open 2021. In her first match back after a hiatus of almost six months, she beat Belinda Bencic in straight sets 6-4 6-1.

Her second round opponent is Sloane Stephens. Stephens is a former Charleston Open Champion, having won it in 2016. She is also a former US Open Champion, 2017, and has been ranked as high as no.3 in the World. At the start of the year, Stephens lost her opening round encounter in four consecutive tournaments  before finally winning her first round match at Miami Open. Her first round opponent here was Wang Xinyu who she beat in straight sets, 6-2 6-4.
